The Flow Cytometry Core Laboratory provides access to state of the art instrumentation and professional flow cytometry services to members of the research community of The Children's Hospital of Philadelphia and University of Pennsylvania; investigators from outside the campus are welcome to our facility. The lab is located in suite 1207 on the 12th floor of the Leonard and Madlyn Abramson Pediatric Research Center. The staff has the required expertise for performing a variety of flow cytometry applications, including sample processing for surface and intracellular staining, functional assays, complex multi-color flow cytometry analyses and cell sorting.

New CytoFLEX LX cytometer
The CytoFLEX LX cytometer has become operational in the flow lab since mid-September, 2017. CytoFLEX has 6 lasers and 21 fluorescence detectors with excellent sensitivity and linearity and it is equipped with a plate loader that can handle 96 and 384 well plates. Microparticles as small as 200 nm can be detected on CytoFLEX by triggering on the FSC detector.
A presentation of the instrument will take place on September 11, 2017 in Abramson Research Center, Room 1210 at 3:30 PM. Everyone interested in polychromatic flow analyses, in high-throughput flow assays or in microparticle studies is invited to join.
FACSCalibur has been relocated
FACSCalibur has been moved on August 15, 2017 in room 1214C in Abramson building. The new location is on the same floor with the Flow Cytometry Lab, two doors down the hallway. FACSCalibur will remain in this location until further notice.

FlowJo licenses are now available at a cost of $275/year to users with a CHOP email address. Each license gives access to FlowJo to one computer (PC or Mac). To request a license fill out this online form. Starting in July we will we will organize small group FlowJo training sessions. New UV laser on LSR II
The UV (355 nm) laser on LSR II has been replaced on May 15, 2017. Older experiment templates may be used but minor adjustments for PMT voltages may be required.
FACSJazz minor configuration change
On Friday, May 5th, 2017 the FACSJazz optical configuration has been modified to accommodate the detection of PE-Cy7. The 710/50 bandpass filter previously installed on a detector off the blue laser has been replaced with a 670nm longpass filter. Please contact the staff if you have questions about this modification.
Cancelation policy change
Starting on May 1, 2017 cell sorting reservations during peak hours (9 AM to 6 PM) must be canceled at least 48 hours prior to the beginning of the scheduled appointment in order to avoid a full charge.
The end users who cannot be sure that their samples will be ready at a specific date and time are encouraged to learn how to operate a sorter of their choice in unassisted mode and schedule the sorts during off peak hours or during weekends. Only fully trained end users are allowed to use sorters in unassisted mode.
Reservations on analyzers must be canceled 24 hours in advance in order to avoid a penalty charge.
The compressor for MoFlo Astrios astrios is now located in a closet adjacent to the main lab
On April 26, 2017 the compressor used with MoFlo Astrios has been relocated in a closet next to the main lab in order to decrease the noise level while the sorter is running. End users who shut down Astrios after evening sorts are requested to turn off the compressed air supply using the valve located on the right side of the instrument. (It is not necessary to turn off the compressor.)
MoFlo Astrios, FACSJazz and Accuri C6 are located in the same room
At the beginning of April 2017 the dividing wall between rooms ARC 1207 A and 1207 B has been removed. MoFlo Astrios and FACSJazz are now in a common space allowing the staff to monitor the sorters more efficiently. Accuri C6 is also located in room 1207 A.
